Course Description

Cell culture has become one of the major tools used in science today. This introductory course aims to discuss the fundamentals of a cell culture laboratory. Participants are expected to gain a comprehensive knowledge on cell biology and laboratory procedure including the needed conditions and techniques to maintain the cell lines. Attendees will learn about interpreting the microscopic images and making conscious decisions on maintaining cell lines. This course offers an overview of cell-based assays today, advantages and outcomes. Moreover, novel cell experiment strategies will be discussed.

Day 1

  • History and introduction of cell culture
  • Equipment of a cell culture laboratory
  • Aseptic techniques 
  • Cell culture types (primary and secondary cell cultures)
  • Development of growth curves

Day 2

  • Observing media (Composition, preparation and changing of media)
  • Plating cells from frozen stocks and freezing cells 
  • Adherent and suspension cell growth
  • Passage/ subcultivation
  • Inverted microscope (cell image interpretation)

Day 3

  • Cell counting via Hemocytometer
  • Viability/ cytotoxicity tests: Trypan Blue, LDH Assay, MTT Assay, Alamar blue
  • Fluorescence Activated Cell Sorting (FACS)

Day 4

  • Cell banks and ordering procedure 
  • Types of biological contamination and prevention 
  • Transport studies (Cell signaling and Teer measurement) 

Day 5

  • Uptake and release studies
