Course Description
A western blot is a biochemical technique with which a specific protein in a mixture of proteins can be detected by means of antibodies. Enzyme-Linked Immuno Sorbent Assay (ELISA) is a quantitative measurement method based on investigating the antigen-antibody relationship and the activity of an enzyme bound to an antibody. This course aims to give the participants theoretical and practical knowledge about Western Blot and ELISA used for different purposes in the R&D laboratory.

Day 1

- Preparation of samples
- Isolation of proteins from cells and tissues
- Spectrophotometry
- Measurement of proteins by Bradford method
Day 2
- Introduction to ELISA
- Preparation solutions of ELISA
- Direct, indirect and sandwich ELISA
- Determination of the concentration with ELISA
- Data analysis and troubleshooting
Day 3
- Introduction to SDS-page gel electrophoresis and equipment
- Calculation procedures
- Preparation of buffers used in SDS-page gel electrophoresis
- Preparation of gels for SDS-page
- Determination of proteins by gel staining (Coomassie blue)
Day 4
- Introduction to Western Blot and equipment
- Preparation of buffers used in Western Blot
- Western Blot protein transfer
- Transferring proteins to the membrane
- Incubation with membrane blocking and primary antibody
Day 5
- Incubation with secondary antibody
- Introduction to microscopy
- Protein detection and visualization image acquisition with a microscope
- Western Blot troubleshooting
